Study name: Effectiveness and safety of Upadacitinib in Real-World clinical practice: results from the national observational study – value of achieving minimal disease activity

Study type: Prospective, Multicenter, Observational study

Patient cohort: Adults and adolescents (≥12 years of age) with moderate to severe AD

Location: 125 locations

Treatment: RINVOQ 15 mg or 30 mg once daily

Study period: Ongoing analysis from December 13, 2021 to November 2026 (Estimated)

Number of patients analyzed: 259

TREATMENT CONSIDERATIONS:

RINVOQ 15 mg once daily is the recommended dose for patients at higher risk of VTE, MACE, or malignancy, adolescents 12 to 17 years of age weighing at least 30kg, and patients ≥65 years of age. If the adolescent patient does not respond adequately to 15 mg once daily, the dose can be increased to 30 mg once daily. The lowest effective dose to maintain response should be used.1

*UP-TAINED is an ongoing prospective, multicenter, observational study in patients with moderate-to-severe AD treated with Upadacitinib 15 or 30 mg daily. Effectiveness and safety are assessed over 2 years. Reported here are 1-year results from an interim analysis. This analysis includes patients completing one year of observation under continuous Upadacitinib treatment. Furthermore, patients discontinuing the study within the first year were included. The results are reported for all included patients as observed without imputation.2

DATA LIMITATIONS: RWE is collected outside of controlled clinical trials and has inherent limitations including a lesser ability to control for confounding factors. Only known confounders were accounted for in the analysis. Inclusion criteria differ from those used in clinical trials. Data are descriptive in nature. Causal inferences and conclusions cannot be made. Limitations associated with observational studies should be considered when interpreting these findings such as the potential for responder bias. Findings from real-world analyses should not be directly compared with those from RCTs. No safety data were collected in this study.

§§ELEVATE: AbbVie defines "ELEVATE" as the raising of the standards of care in AD patients, measured by WP-NRS and EASI score, treated with Dupilumab vs RINVOQ at week 16 of the head to head trials.4,5
